82P Correlations of androgen receptor (AR) and transforming growth factor beta receptor type 2 (TGFbR2) expression in prostate cancer (PCa)

Transforming growth factor beta (TGFb) is a potent inducer of epithelial-mesenchymal transition. Of all members the TGFb receptor family only TGFbR2 capable direct binding to ligand. In experimental conditions interactions were shown between and AR signaling pathways, often leading negative feedback loops. However, data on expression levels both receptors in clinical cancer samples are scarce. was studied 31 radical prostatectomy specimens. Double immunofluorescence used with primary mouse monoclonal anti-AR (BioGenex, 1:100 dilution) rabbit polyclonal anti-TGFbR2 (elabscience, 1:100) antibodies (Ab) secondary goat Ab (ThermoFischer, 1:200) labelled Alexa Fluor 488 555. 3-12 random high-power fields (HPF) (x400) per case (total 277 HPFs) studied, staining characteristics assessed semi-quantitatively (intensity: 1 – weak, 2 moderate, 3 strong; proportion stained cells: 0 absent, up 1/3, 1/3 2/3, more than 2/3). nuclear seen mostly PCa cells.
